is it possible to use two 72hr transfer passes within 3 weeks to the same city

Im planning UK to Shanghai - Tokyo - Shanghai - London 2/3 weeks in Japan

Yes, and actually you are actually eligible for 144-hour visa free transit in Shanghai. The 72-hour has been extended to 144-hour years ago.
