public transport from HK airport to Best Western Plus Hotel, Kowloon.

Is it ok to have suitcase to bring into the public bus?
Is there a direct bus from the airport to the hotel?

Yes, it is okay to bring your suitcase into the public bus.

Yes, you can take the Bus Line A12 directly from the airport to Water Street. It takes about 1 hour to reach. Get off the bus and walk about 90m to your hotel.
