About:240-Hour Visa-Free Transit

Hong Kong- Shenzhen by bus

I'm traveling from Boston to Hong Kong. I would like to visit Shenzhen by bus for three days. Can I still apply the 144 hours visa without stop by Guangdong airport?

No, this is not possible. The only option to travel to Guangdong province from Hong Kong, using any kind of visa free transit scheme, is to fly from Hong Kong to Guangzhou. Of course, you can rely on 5 day Shenzhen VOA, although it is quite expensive for US citizens and also on 6 day visa exemption for tourists to Pearl River Delta. This options has to be organized through a Hong Kong based and authorized tour agency and if the itinerary and the price are suitable, you can travel to very wide area of Guangdong province, Shenzhen included, completely visa free.
