No, it is not possible to do that in the way you have described.
In order to obtain 144 hours visa free transit you have to:
1. Fly from Hong Kong to Guangzhou. There is no other way to arrive in Guangdong, but by flying in order to be eligible for 144 hours visa free transit.
2. Stay within Guangdong. The entire province is allowed for free movement
3. Take a ferry from Shenzhen (Shekou port) to Macau and then you can return from Macau to Hong Kong. It is required to have ferry ticket before you board the plane in Hong Kong for Guangzhou and you can buy this ticket on TurboJet website.
As you can see, you have to make a real transit and it means: Hong Kong - China (Guangdong province) - Macau. Any other combination is non valid. You will apply for your 144 hours visa free transit upon arrival at Guangzhou airport.
