We are staying at the Wyndham Grand Plaza 2288 Pudong Ave, where would we pick up the hop on hop off bus please? Would we need a taxi to the nearest pick up point?

Yes, you need to take a taxi to Jinmao Tower, the nearest bus stop for the sightseeing bus. Btw, the taxi takes about 15mins and CNY 25 or so.
