About:72-Hour Visa-Free Transit

Do I need a visa if I have two transfer stops in China,

namely first in Guangzhou (CAN) and then in Peking (PEK)? The schedule is:

8:40 AM MEL -> 3:20 PM CAN
7:00 PM CAN -> 10:10 PM PEK
00:30 AM PEK -> 4:40 AM AMS

I have booked all three flights as 1 booking/ticket.

You need a visa. Transit incl. multiple stops within China (People's Rep.) is permitted at Guangzhou (CAN) only when the first transit point is Beijing (PEK). The total maximum transit time is 24 hours.
