About:72-Hour Visa-Free Transit

I'm flying into Beijing from Korea (US prior to that) , flying out the same day to Shenzhen,

spending a night in Shenzhen and flying out to US. Does the 72 hr visa work for this itinerary?

No. You can't have 72 hours free transit if you have two stops in China.
