Just set off early and one full day shall be enough! I suggest you visit the Buddha first. Afterwards, you can spend the whole afternoon till night at the resort.

Anyway, from there to Giant Buddha, take Disneyland Resort Line to Sunny Bay Station and Tung Chung Line to Tung Chung Station, Exit A. Then walk to Lower Cable Car Terminal and take it to the destination. The return way is the same.

From Giant Buddha to your hotel, you can go back to Tung Chung Station first. Then take Tung Chung Line to Nam Cheong Station and transfer to West Rail Line to East Tsim Sha Tsui Station, Exit N5. After arriving, walk northwest around 120 yards to find Tsim Sha Tsui Station.

1. Yes, you are right.
2. After visiting the resort, you may take Disneyland Resort Line, get off at Sunny Bay Station. Next, transfer to Tung Chung Line, and get off at Nam Cheong Station. Then, transfer to West Rail Line, and get off at East Tsim Sha Tsui Station. The fare is HKD 19.8, and the duration is 40 minutes. :)
