About:Visa Policy of China

Am I eligible for a 72 hour transit visa?

I hold a USA passport and will be traveling to Shanghai from Hong Kong and then flying to Macau 2 days later.

Well, your route is ok. However, I am afraid that the immigration might stop you because they think you intentionally make use of this policy to avoid applying Chinese entry visa. There is a big risk that you might be stopped. In your case, I think you should apply for a L type in HK before you go to Beijing, to play it safe.
