About:Macau Visa Policy

What are requirements for travel to Hong Kong to Macau to Shenzhen from/to USA?

If I have a China Visa that allows 2 entries in 6 months, Can I fly into Hong Kong, travel to Macau and then to Shenzhen before returning to Hong Kong for return flight to USA?

You are not required visa to both HK and Macao, so a single entry Chinese visa is enough if you will fly from USA to HK then travel as the itinerary you listed above.
