I am an US citizen living in China for more than 2 years with an annual L-visa. I am also married to a Chinese National for more than 2 years.I now have all documents for a Work visa applications. Can I apply for the Z-visa in Shenzhen China Entry Office?

No, you need to apply for a China work visa outside of mainland China. You can try to apply in Hong Kong through the Office of the Ministry of China Foreign Affairs.
