The coach from Guangzhou to Nanjing will take you at least 26 hours on the way. You'd better fly to Nanjing.

I recommend the overnight sleeper train from Hong Kong or Guangzhou to Shanghai, and transferring to a high speed train to Nanjing, all in under 15 hours. A bus wouild be highly uncomfortable.
