First of all, you have to apply for a China visa to visit Shenzhen. Although China visa on Arrival(https://www.travelchinaguide.com/embassy/visa/service.htm) to Shenzhen is issued to foreign travellers, you could only get this kind of visa in Hong Kong(https://www.travelchinaguide.com/cityguides/hongkong/). So, if you would like to visit Shenzhen(https://www.travelchinaguide.com/cityguides/guangdong/shenzhen/), you must transfer at Hong Kong. From Macau, you have to take a ferry to downtown Hong Kong firstly. Then, from Hung Hom stattion, Kowloon, you have to take metro of Hong Kong to the Luohu Border. Here, you could apply for the onsite visa to enter into Shenzhen.
Ps: When return to Macau, you could take a ferry at Shenzhen Shekou Port or transfer in Hong Kong again.

Macau is not large so that you could see these main sightseeings in Macau(https://www.travelchinaguide.com/attraction/macau/) in 2 days. The tourist spots here are full of Portuguese flavor, which is totally different from Shenzhen. In fact, Shenzhen is a modern and industrial city, a good profile of developing mainland China. Here, you could travel around Luohu district, where you will find some famous parks (Window of the World, Splendid China), large wholesale markets and local dim sum. If you are interested, 2 days will be ok for a Shenzhen Tour(https://www.travelchinaguide.com/package/shenzhen.htm).
