Hangzhou to Wuzhen: go to Hangzhou East Bus Station to take direct bus to Wuzhen. It costs around CNY 25 and takes more than one hour.

Hangzhou to Zhouzhuang: take bus to Suzhou South Bus Station from Hangzhou East Bus Station. Then take bus from Suzhou South Bus Station to Zhouzhuang. Or you can go to Kunshan first and then take bus to Zhouzhuang.

CNY25 is just bus ticket fare. After arriving at Wuzhen, you need to pay entrance fee. Check this page to learn the entrance fee.

If I were you, I will choose Wuzhen instead of Zhouzhuang. Zhouzhuang might be more famous but it has been too commercialized. If you plan to experience real and orginal water town, better go to Wuzhen.
