You'll need a visa for China.

Sorry, my question is whether I can use the 144 hour visa-free entry in my case.

Your travel route is Brisbane - Singapore - Shanghai - Singapore - Brisbane. Thus you are not eligible for the 144 hours visa free policy.

It obviously does not qualify for 144 hours transit without visa because it’s a visit to China and not a transit! The rules are quite straightforward and clear!
