Is there a train with luxury seats (private seats with own toilet) running from Dunhuang to Shanghai and if available, how long does it take?

Actually, the train from Dunhuang to Shanghai is not recommended as it takes around 32 to 37 hours which is too long. You may feel exhausted by taking train for such a long time. Besides, there is no luxury seats available on the train. Therefore, you are suggested to take flight from DH to SH instead.
