About:144-Hour Visa-Free Transit

Can I travel from US to Shanghai (transiiting/changing plane in Japan), then to Japan?

Sorry, you can't. You need to take a direct flight from US to Shanghai or a flight that stops over other countries before arriving in Shanghai to make yourself eligible.
