You are not eligible for the 144 hour free transit because you stop in two Chinese cities. But if your total layover time in Shanghai and Beijing is less than 24 hours, you can use the direct transit and no visa is required. Otherwise, you need to apply for a transit G type or tourist L type ahead of time.

Cococ, if you want to use the free transit, you cannot travel like that.
The 144 hour policy is only carried out in Shanghai, Zhejiang and Jiangsu.
In Beijing, only 24 hour direct transit and 72 hour free transit are available. But as for the 72 hour free transit, you are only allowed to stop in ONE Chinese city.
According to what you said, you need to stop in two Chinese cities and the total layover time is more than 24 hours, so you jhave to apply for a transit G type or tourist L type ahead of your journey!

Coco, if you fly to Beijing from USA and then fly to HK or Macau, you can enjoy the 72 hour free transit in Beijing.
After, if you fly to Shenzhen for less than 24 hours from HK or Macau and then fly back to USA, you can enjoy the 24 hour direct transit in Shenzhen. In this case, no visa is required for your travel.
But if you will stay for more than 24 hours in Shenzhen, a visa is needed.

Coco, if you go to Shenzhen from HK and then go back to HK, you need to apply for a visa to visit Shenzhen.
You can travel to Macau from HK because US passport holders can enjoy free entry to Macau for a stay of no more than 30 days.
