what is the mode of transport, perferrable by bus, from Macau border and reach HK airport and catch a flight by 5.00am on saturday.

I'm afraid no public transportation is available that early! Therefore, if you don't wanna take a taxi or rent a car to get there, I suggest you go to HK one day before to stay there for a night and catch your early flight!
